چکیده مبسوط
مقدمه و هدف: استفاده از بافرهای شیمیایی از جمله بیکربنات­ سدیم و افزودنی باکتریایی نظیر مگاسفرا السدنی باعث بهبود هضم و تخمیر جیره­ ی با کنسانتره بالا می­شود. لذا، این پژوهش با هدف مقایسه‌ی تاثیر استفاده از باکتری مصرف­ کننده اسید و بافرهای مختلف شیمیایی برای بهبود قابلیت هضم و تخمیر جیره‌های پرکنسانتره و یافتن بافر مناسب و مقرون به صرفه انجام شد.
مواد و روش‌ها: از چند بافر شیمیایی به تنهایی یا همراه با باکتری مگاسفرا السدنی به عنوان بافر بیولوژیکی استفاده شد. در این آزمایش از روش‌های هضم­ دو مرحله­ ای، کشت اختصاصی قارچ­ها و باکتری­های بی­ هوازی شکمبه استفاده شد. تعداد ۱۲ تیمار شامل ۱- جیره‌ی شاهد (فاقد افزودنی و حاوی 70 درصد کنسانتره + 30 درصد علوفه یا جیره‌ی پایه)، ۲- جیره‌ی پایه+ ۳ میلی‌لیتر باکتری مگاسفرا السدنی (cfu/ml 10۸ × 1/5)، ۳ تا ۱۲- یک درصد از ۵ بافر بنتونیت سدیم،  بیکربنات سدیم، اکسید منیزیم، زئولیت، سدیم سسکوئی کربنات به تنهایی یا همراه با باکتری مگاسفرا السدنی بودند.  
یافته‌ها: در آزمایش هضم دو مرحله ­ای، قابلیت هضم مادهی خشک در تیمارهای دریافتکنندهی بافر و باکتری  بهبود یافت (0/05>p). قابلیت ­هضم ماده خشک، الیاف نامحلول در شوینده خنثی و الیاف نامحلول در شوینده اسیدی توسط قارچ ­های جداشده از شکمبه تحت تاثیر تیمارهای آزمایشی قرار گرفت و در تیمار حاوی بافر شیمیایی + باکتری بیشتر از شاهد بود (0/05>p). استفاده از بافر شیمیایی و بیولوژیکی تاثیری  بر قابلیت هضم مواد مغذی توسط باکتری‌های شکمبه  داشت و در تیمارهای حاوی بافر + باکتری بیشتر از شاهد بود (0/05>p). فراسنجه ­های تخمیری محیط کشت باکتری و قارچ‌های شکمبه شامل pH و غلظت نیتروژن آمونیاکی  به‌طور معنی‌داری تحت تاثیر تیمارهای آزمایشی قرار گرفتند و به ترتیب در همه‌ی تیمارها بیشتر و کم‌تر از تیمار شاهد بودند (0/05>p).
نتیجه‌گیری: استفاده از بافر شیمیایی باعث بهبود شرایط هضم و تخمیر جیره‌های پر کنسانتره بره‌های پرواری شد و در بسیاری از موارد اثر باکتری مصرف کننده‌ی اسید با بافرهای شیمیایی قابل رقابت بود. از طرفی، استفاده‌ی توام باکتری و بافر نیز اثرات هم‌افزایی داشتند.
