fa طراحی، ساخت و بیان ‌اپی‌توپ‌های پروتئین VP1 ویروس تب برفکی سروتیپO ایران Design and Construction of Multiple-Epitope Recombinant Vaccine Against Foot-and-Mouth Disease Virus Type O تخصصي Special پژوهشي Research تب برفکی (FMD)، یک بیماری حاد بسیار مسری در حیوانات زوج سم است و باعث زیان‌های اقتصادی قابل توجه‌ای میشود. بنابراین، ساخت یک واکسن امن‌تر و کارآمد‌تر در برابر ویروس تب برفکی (FMDV) ضروری است. هدف از این مطالعه توسعه یک واکسن کاملاً ایمن چند اپی‌توپی نوترکیب به جای واکسن‌های سنتی غیرفعال، در برابر ویروس  (FMDV)تب برفکی O ایران بود. برای هم‌سانه‌سازی و بیان ‌اپی‌توپهای نوترکیب ایمنی‌زا بر علیه ویروس تب برفکی، ژن پروتئین نوترکیب متشکل از ترکیبات مختلف اپی‌توپ‌های سلول‌های B و سلول‌های T از پروتئینVP1  کپسید ویرس FMD سنتز شد. قطعه 1155 جفت بازی ژن نوترکیب با روش PCR با استفاده از آغازگرهای اختصاصی تکثیر و محصول PCR خالص و در ناقل بیانی pET-32A کلون شد. بیان پروتئین نوترکیب، با استفاده از یک میلیمولار ایزوپروپیل -بتا-D-گالاکتوزید (IPTG) القا و تأئید بیان پروتئین نوترکیب تولید شده با استفاده از الکتروفورز پروتئین SDSPAGE و دات بلاتینگ انجام گرفت. صحت کلونینگ ژن پروتئین نوترکیب چند اپی‌توپی با روش PCR و هضم آنزیمی و تعیین توالی تأئید شد. تجزیه و تحلیل SDS-PAGE نشان داد که سیستم بیانی پروکاریوتی pET32a-RP تولید پروتئین نوترکیب هدف، با وزن مولکولی در حدود 60 کیلو دالتون کارایی دارد و پروتئین نوترکیب چند اپی‌توپ به صورت نامحلول بیان شد. نتایج نشان داد سیستم بیانی پروکاریوتی جهت تولید ساختارهای چند اپی‌توپی نوترکیب ایمنی‌زای ویروس تب برفکی مؤثر و می‌تواند به عنوان یک روش بالقوه برای سازههای پلیاپیتوپها استفاده شوند.      Foot-and-mouth disease (FMD) is an extremely infectious and occasionally fatal viral disease with a rapid onset and a short course that affects cloven-hoofed animals and results in considerable financial losses. For this reason, availability of more reliable and efficient vaccines against Foot-and-mouth disease virus (FMDV) is felt as a necessity. The purpose of this study is to develop a perfectly safe immunogen in order to supplant the usual inactivated vaccine. A tandem repeat multiple-epitope recombinant vaccine against foot-and-mouth disease (FMD) virus (FMDV) type O was developed. In order to develop recombinant epitope vaccines against foot-and-mouth disease virus (FMDV), multiple-epitope recombinant vaccine (rP) comprising various combinations of B cell and T cell epitopes from VP1 capsid protein were synthesized. A 1155-bp gene fragment was amplified by PCR using specific primers. The amplicon was purified and then cloned into expression vector pET-32a. For expression of recombinant protein, pET32a-rP plasmid was transformed into Escherichia coli BL21 (DE3) cells. Recombinant protein was overexpressed with isopropythio-beta-D-galctoside (IPTG). SDSPAGE and DOT blotting were performed for protein determination and verification.The multiple-epitope recombinant was validated by colony-PCR and enzymatic digestion. IPTG with a dosage of 1.0 mmol/L could effectively promote protein expression.  SDS-PAGE analysis demonstrated that our constructed prokaryotic expression system pET32a-rP efficiently produces a target recombinant protein with a molecular weight of about 60 kDa.
