fa بررسی اثر ترکیبات غیر استروییدی (عصاره پوست درخت بید، بره موم و داروی ضد التهاب سلوکسیب) بر بیان برخی از ژن های پیش التهابی، ضد التهابی و فاکتورهای رشد شبه انسولین در مرغ های مادر گوشتی مسن Studying the effect of non-steroidal compounds (willow bark extract, propolis, and the anti-inflammatory drug celecoxib) on the expression of some pro-inflammatory, anti-inflammatory, and insulin-like growth factors genes in aged broiler breeders فیزیولوژی فیزیولوژی پژوهشي Research مقدمه و هدف: با افزایش سن مرغهای تخم‌گذار، تغییرات ریزساختاری فولیکول‌ها همراه با علامت‌های التهاب و استرس اکسیداتیو گزارش شده است. عوارض التهابی مرتبط با سن در مرغهای مادر گوشتی می‌تواند با ایجاد عدم تعادل میان ژن‌های پیش‌التهابی (مانند IL-1β، IL-6، TNF-α و COX-2) و ژن‌های ضدالتهابی (نظیر IL-10 و TGF-β)، کارایی تولیدمثلی، کیفیت تخم و وضعیت متابولیکی را مختل کند. در سال‌های اخیر، سه رویکرد غیر‌استروئیدی برای تعدیل التهاب و بازآرایی بیان ژن‌های مرتبط مورد توجه قرار گرفته‌اند: عصاره‌ی پوست درخت بید (حاوی سالیسین)، بره‌موم و داروی ضدالتهاب انتخابیِ COX-2 ( سلوکسیب). هدف ازاین پژوهش براین است که این سه رویکرد بتوانند محرک تولید فاکتوهای ضد التهابی شده و منجر به کاهش و کنترل التهاب در اواخر دوره تخمگذاری در مرغ مادر گوشتی شود. مواد و روشها: برای انجام این پژوهش تعداد ۸۰ قطعه مرغ و ۱۰ قطعه خروس نژاد راس (سن مرغهای مادر ۵۲ هفته با میانگین وزن ۳۸۶۰) در سالن پرورش دانشکده کشاورزی و منابع طبیعی ساری به مدت ۵ هفته (هفته اول سازگاری و چهار هفته آزمایش تیمارها) در قالب طرح کاملا تصادفی با چهار تیمار و چهار تکرار نگهداری شدند. تیمارهای آزمایشی شامل: تیماراول: شاهد با تغذیه جیره پایه، تیمار دوم: جیره پایه به علاوه عصاره الکلی بره موم با دوز ۱۰۰ میلیگرم به ازای وزن متابولیک بدن هر مرغ در روز به صورت خوراکی به مرغها داده شد. تیمارسوم: جیره پایه به علاوه داروی ضد التهاب (غیر استروییدی) سلوکسیب با دوز ۱۵ میلیگرم به ازای وزن متابولیک بدن هر مرغ به صورت خوراکی به مرغها داده شد. تیمار چهارم: جیره پایه به علاوه عصاره الکلی پوست درخت بید با دوز ۱۵۰ میلیگرم به ازای وزن متابولیک بدن هر مرغ در روز به صورت خوراکی به مرغها داده شد. در روز پایان پژوهش ازیک قطعه مرغ از هر تکرار cc۲ خون از سیاهرگ بال گرفته شد. نمونهها در منهای۸۰ درجه سانتیگراد تا زمان انجام مراحل سنجش بیان ژن نگهداری شدند. ژن ها شامل  IGFBp2, IL-1,  IL-6, IL-10, IGF-1, IGF-2, IGF-1R و TNF-α بود. بیان نسبی ژنها به وسیله واکنش Real-Time PCR با استفاده از آغازگرهای اختصاصی ژنها اندازهگیری شد. داده های بدست آمده از بیان ژن با روش لیواک واکاوی اولیه شد. واکاوی دادهها با رویهGLM نرمافزار SAS محاسبه شد. یافتهها: در نتایج این پژوهش کاهش معنیدای در بیان نسبی ژن IGF-2 در تیمار ۳ مشاهده شد (P<0/05). همچنین کاهش یا افزایش معنیداری در بیان نسبی ژنهای مورد بررسی بین کلیه تیمارها دیده نشد. (P<0/05). نتیجه‌گیری: احتمالا بهواسطه سن بالای مرغها ، التهاب بالا و استرس غیر قابل کنترل محیطی، دوزهای مصرف شده نتوانست باعث کاهش التهاب و اثر معنیداری بر بیان نسبی ژنهای مورد آزمایش بگذارد. احتمال دارد با افزایش دوز عصارهها، داروی ضد التهاب (سلوکسیب) و همچنین افزایش مدت آزمایش، اثر معنی داری در بیان نسبی ژنهای مورد آزمایش دیده شود.   Introduction and Objective: As laying hens age, microstructural changes in follicles have been reported, along with signs of inflammation and oxidative stress. Age-related inflammatory complications in broiler breeders can impair reproductive performance, egg quality, and metabolic status by creating an imbalance between proinflammatory genes (such as IL-1β, IL-6, TNF-α, and COX-2) and anti-inflammatory genes (such as IL-10 and TGF-β). In recent years, three nonsteroidal approaches to modulate inflammation and reprogram the expression of related genes have received attention: willow bark extract (containing salicin), propolis, and the selective COX-2 anti-inflammatory drug (celecoxib). The aim of this study is to determine whether these three approaches can stimulate the production of anti-inflammatory factors and lead to the reduction and control of inflammation in aged broiler breeders. Material and Methods: To conduct this study, 80 hens and 10 roosters of the Ross breed (the age of the hens was 52 weeks with an average weight of 3860) were kept in the breeding hall of the Faculty of Agriculture and Natural Resources of Sari for 5 weeks (the first week was for adaptation and 4 weeks for treatment testing) in a completely randomized design with 4 treatments and 4 replications. The experimental treatments included: The first treatment: control with basal diet, the second treatment: basal diet plus 100 mg of propolis alcoholic extract per metabolic body weight of each hen per day was given orally to the hens. The third treatment: basal diet plus the anti-inflammatory drug (non-steroidal) celecoxib at a dose of 15 mg per metabolic body weight of each hen per day was given orally to the hens. The fourth treatment: basal diet plus 150 mg of willow bark alcoholic extract per metabolic body weight of each hen per day was given orally to the hens. On the last day of the study, 2cc of blood was collected from the wing vein of one chicken from each replicate. The samples were stored at -80°C until the gene expression assay was performed. The genes included: IGFBp2, IL-1 IL-6, IL-10, IGF-1, IGF-2, IGF-1R, and TNF-α. The relative expression of the genes was measured by Real-Time PCR using gene-specific primers. The data obtained from gene expression were initially analyzed by the Livak method. Data analysis was calculated using the GLM procedure of SAS softwar Results: In the results of this study, a significant decrease in the relative expression of the IGF-2 gene was observed in treatment 3 (P<0.05). Also, no significant decrease or increase in the relative expression of the genes studied was observed between all treatments (P<0.05). Conclusion: Probably due to the advanced age of the chickens, high inflammation and uncontrolled environmental stress, the doses used failed to reduce inflammation and have a significant effect on the relative expression of the tested genes.
