مقدمه و هدف: افزایش نگرانی نسبت به مقاومت&#8204; های ضدمیکروبی و تبعات استفاده بلند مدت از آنتی&#8204;بیوتیک&#8204; ها در صنعت طیور، توجه را به جایگزین&#8204; های طبیعی و ایمن جلب کرده است. دو راهکار پر کاربرد در این زمینه، افزودنی&#8204; های گیاهی سرشار از ترکیبات زیست&#8204; فعال و پروبیوتیک&#8204; ها هستند که می&#8204; توانند با بهبود تعادل میکروبی روده، تقویت وضعیت اکسیداتیو و ارتقای کارایی گوارش، سلامت و بازده تولید را ارتقا دهند. گیاه خرفه (&lt;em&gt;Portulaca oleracea&lt;/em&gt;) به&#8204;سبب دارا بودن اسید های چرب غیر اشباع امگا-۳، فلاونوئید ها و پلی&#8204;فنول&#8204; ها، ویتامین&#8204; ها و املاح، خواص آنتی&#8204; اکسیدانی، ضد التهابی و ضد میکروبی دارد. از سوی دیگر، مخمر &lt;em&gt;saccharomyces&lt;/em&gt; &lt;em&gt;cerevisiae&lt;/em&gt;&amp;nbsp; به&#8204; عنوان مخمر شناخته&#8204; شده با توانایی بهبود تعادل میکروفلور، افزایش هضم و جذب و تحریک ایمنی مخاطی مطرح است. بلدرچین ژاپنی (&lt;em&gt;Coturnix japonica&lt;/em&gt;) به &amp;lrm;دلیل رشد سریع و ضریب تبدیل مناسب، مدل مناسبی برای ارزیابی اثرات تغذیه&amp;lrm; ای این ترکیبات به&#8204; شمار می&#8204;رود. هدف پژوهش حاضر، بررسی اثر سطوح مختلف پودر بذر خرفه و مخمر &lt;em&gt;S. cerevisiae&lt;/em&gt; بر عملکرد تولیدی،کیفیت تخم، ویژگی های لاشه، pH دستگاه گوارش، جمعیت میکروبی روده، فعالیت آنزیم&#8204; های آنتی&#8204; اکسیدانی و شاخص&#8204; های بیوشیمیایی خون و ریخت&#8204; شناسی روده در بلدرچین بود.&lt;br&gt;
مواد و روش&#8204; ها: آزمایش به&#8204;صورت طرح کاملاً تصادفی با چهار تیمار و پنج تکرار اجرا شد؛ هر تکرار شامل ۱۰ قطعه بلدرچین ژاپنی نر و ماده همسن بود. تیمار ها عبارت بودند از&amp;nbsp; ۱) جیره پایه (شاهد)، ۲( جیره پایه + ۱ درصد پودر بذر خرفه، ۳) جیره پایه + ۲ درصد پودر بذر خرفه، و ۴) جیره پایه + 0/5 درصد مخمر &lt;em&gt;S. cerevisiae &lt;/em&gt;جیره&#8204;ها مطابق توصیه&#8204;های NRC (1994) به&#8204;صورت ایزوکالریک و ایزونیتروژن فرموله شدند. دوره پرورشی ۴۲ روز بود. در پایان دوره، شاخص&#8204; های عملکردی شامل وزن نهایی، افزایش وزن روزانه، و مصرف خوراک ثبت شدند و ضریب تبدیل غذایی محاسبه شد. برای ارزیابی اجزای لاشه، پس از کشتار، درصد لاشه، کبد، قلب، چربی بطنی و سنگدان در دو پرنده از هر تکرار اندازه&#8204;گیری شدند. نمونه&#8204; برداری از محتویات روده برای شمارش &lt;em&gt;Escherichia coli&lt;/em&gt; و .&lt;em&gt;Lactobacillus&lt;/em&gt; spp&amp;nbsp; انجام گرفت و pH محتوای روده با pH متر دیجیتال اندازه&#8204;گیری شد. وضعیت اکسیداتیو و بیوشیمی خون با سنجش مالون&#8204; دی&#8204; آلدئید، فعالیت سوپراکسید دیسموتاز و گلوتاتیون پراکسیداز، و نیز کلسترول تام، تری&#8204;گلیسرید، HDL، LDL و آنزیم های کبدی ارزیابی شد. برای بررسی ریخت&#8204; شناسی، مقاطع بافتی دوازدهه و ایلئوم رنگ&#8204; آمیزی&#8204; شده با هماتوکسیلین&amp;ndash;ائوزین تهیه و ارتفاع پرز، عمق کریپت و نسبت پرز به کریپت اندازه&#8204; گیری گردیدند. داده&#8204;ها با نرم&#8204;افزار SAS و آزمون دانکن در سطح ۵ درصد مقایسه شدند.&lt;br&gt;
یافته&#8204; ها: افزودن ۲ درصد پودر بذر خرفه، کاهش معنی&#8204; دار شمارش ای. کولای را تا حدود ۱۶۰۰ واحد و افزایش چشمگیر لاکتوباسیلوس&#8204; ها را تا حدود ۲۳۵۰۰ واحد نسبت به شاهد رقم زد. تیمار حاوی مخمر &lt;em&gt;S. cerevisiae&lt;/em&gt; نیز کاهش قابل&#8204; توجه ای. کولای و افزایش لاکتوباسیلوس&#8204; ها را نشان داد، هر چند که شدت اثر آن از تیمار ۲ درصد خرفه کمتر بود. ارزیابی اجزای لاشه نشان داد که افزودن خرفه و مخمر بر درصد لاشه و اغلب اندام&#8204; ها اثر معنی&#8204; داری نداشت؛ با این&amp;lrm;حال، درصد کبد در تیمار ۲ درصد خرفه نسبت به شاهد به&#8204; طور معنی&#8204;دار کاهش یافت که می&#8204;تواند منعکس&#8204; کننده تعدیل بار چربی و بهبود وضعیت متابولیکی باشد. pH محتوای روده در تیمار های دریافت&#8204; کننده خرفه و مخمر کاهش یافت و بیشترین افت pH در تیمار مخمر مشاهده شد؛ وضعیتی که به نفع استقرار باکتری&#8204; های اسید دوست مانند لاکتوباسیلوس&#8204; ها تلقی می&#8204; شود. در سطح سیستمیک، تیمار ۲ درصد خرفه سبب کاهش مالون الدئید و افزایش فعالیت آنزیم های کبدی شد که بیانگر تقویت ظرفیت ضد&#8204; اکسایشی و کاهش پر اکسیداسیون لیپیدی است. همین تیمار باعث بهبود پروفایل چربی خون شد؛ به&#8204; گونه&#8204; ای که کلسترول تام، تری&#8204; گلیسرید و LDL کاهش و HDL افزایش یافت. در بررسی بافت&#8204; شناسی، افزودن ۱ و ۲ درصد خرفه ارتفاع پرز و نسبت پرز به کریپت را در دوازدهه و ایلئوم افزایش و عمق کریپت را کاهش داد؛ شاخص&#8204; هایی که معمولاً با افزایش سطح جذب و کارایی گوارش همبستگی دارند. مخمر نیز اثرات مثبتی بر این متغیرها داشت، اما تیمار حاوی 2 درصد خرفه افزایش معنی&amp;lrm; داری در این شاخص &amp;lrm;ها نسبت به مخمر ایجاد کرد. برآیند این نتایج نشان می&#8204; دهد که هر دو افزودنی قادر به تعدیل محیط روده و تقویت جمعیت&#8204; های مفید هستند، ولی بذر خرفه در سطح ۲ درصد هم&#8204; زمان بهبود های میکروبی، بافتی و بیوشیمیایی گسترده&#8204; تری ایجاد کرد. همچنین، افزودن 2 درصد پودر بذر خرفه موجب بهبود معنی&amp;lrm; دار شاخص &amp;lrm;های کیفیت تخم شامل افزایش واحد هاو، افزایش ضخامت پوسته و بهبود شاخص زرده نسبت به تیمار شاهد شد، در حالی&amp;lrm;که تیمار مخمر نیز بهبود نسبی ایجاد کرد اما معنی&amp;lrm;دار نبود. ترکیبات زیست&#8204; فعال موجود در بذر خرفه&amp;mdash;از امگا‑۳ تا فلاونوئید ها و پلی&#8204;فنول&#8204; ها می&#8204;توانند سازوکار اثرات مشاهده&#8204; شده را توضیح دهند. اسید های چرب امگا‑۳ با تعدیل مسیر های التهابی و بهبود یکپارچگی غشا، همراه با ترکیبات فنلیِ پاک&#8204; کننده رادیکال&#8204; های آزاد، به کاهش استرس اکسیداتیو و حفاظت از اپیتلیوم روده کمک می&#8204; کنند. در چنین بستری، کاهش pH و افزایش اسیدهای آلی تولیدیِ میکروفلور مفید، رقابت را به زیان پاتوژن&#8204; هایی مانند ای. کولای تغییر می&#8204; دهند و جذب مواد مغذی را تسهیل می&#8204; کنند. &lt;em&gt;S. cerevisiae&lt;/em&gt; نیز با تولید متابولیت&#8204;های مفید و بهبود فعالیت آنزیمی روده، پایداری اکوسیستم میکروبی را تقویت می&#8204; کند؛ هرچند که در این مطالعه دامنه اثر آن نسبت به ۲ درصد خرفه محدودتر بود.&lt;br&gt;
نتیجه&#8204; گیری: نتایج نشان می &amp;lrm;دهند که استفاده از پودر بذر خرفه، به&#8204;ویژه در سطح ۲ درصد، با بهبود تعادل میکروفلور مفید، کاهش ای. کولای، افزایش فعالیت آنزیم&#8204; های آنتی&#8204; اکسیدانی، کاهش پراکسیداسیون لیپیدی، اصلاح پروفایل لیپیدی خون و بهینه&#8204; سازی معماری پرزهای روده، وضعیت گوارشی و سلامت عمومی بلدرچین ژاپنی را ارتقا می&#8204; دهد. مخمر &lt;em&gt;S. cerevisiae&lt;/em&gt; نیز در بهبود فلور روده و کاهش pH مؤثر بود، اما در برخی شاخص&#8204; ها از جمله شاخص &amp;lrm;های بافت&#8204; شناسی و بیوشیمیایی از تیمار ۲ درصد خرفه عقب&#8204; تر ماند. بنابر این، بذر خرفه می&#8204; تواند به&#8204;عنوان افزودنی خوراکی گیاهی کارآمد برای بهبود سلامت و کارایی پرورشی بلدرچین&#8204; ها پیشنهاد شود و در کاهش وابستگی به آنتی&#8204;بیوتیک&#8204; ها نقش داشته باشد. پیشنهاد می&#8204; شود که در پژوهش&#8204; های آینده، بررسی سطوح بالاتر یا ترکیب هم&#8204; زمان بذر خرفه با مخمر&#8204; ها و نیز پایش طولانی&#8204; مدت شاخص&#8204; های تولیدی و ایمنی مدنظر قرار گیرد تا دامنه دوز&amp;ndash;پاسخ و پایداری اثرات روشن&#8204; تر شود.&lt;/span&gt;&lt;/span&gt;&lt;/div&gt;</abstract_fa>

Two widely used approaches in this area are phytogenic feed additives rich in bioactive compounds and probiotics, which can improve intestinal microbial balance, enhance antioxidant status, and improve digestive efficiency, thereby promoting health and production performance. Purslane (&lt;i&gt;Portulaca oleracea&lt;/i&gt;) exhibits remarkable antioxidant, anti-inflammatory, and antimicrobial properties due to its content of omega-3 polyunsaturated fatty acids, flavonoids, polyphenols, vitamins, and minerals. On the other hand, the yeast &lt;i&gt;Saccharomyces cerevisiae&lt;/i&gt;, as a well-known probiotic, improves gut microflora balance, enhances digestion and nutrient absorption, and stimulates mucosal immunity. Japanese quail (&lt;i&gt;Coturnix japonica&lt;/i&gt;) is considered a suitable experimental model for evaluating the nutritional effects of these compounds due to its rapid growth and favorable feed conversion ratio. Therefore, the present study aimed to investigate the effects of different levels of purslane seed powder and &lt;i&gt;S. cerevisiae&lt;/i&gt; yeast on the productive performance, carcass traits, gastrointestinal pH, intestinal microflora, antioxidant enzyme activity, serum biochemical indices, and intestinal morphology in Japanese quails.&lt;/span&gt;&lt;/span&gt;&lt;br&gt;

The treatments were (1) a basal diet (control), (2) a basal diet + 1% purslane seed powder, (3) a basal diet + 2% purslane seed powder, and (4) a basal diet + 0.5% &lt;i&gt;S. cerevisiae&lt;/i&gt; yeast. Diets were formulated to be iso‑caloric and iso‑nitrogenous according to NRC (1994) recommendations. The experimental period lasted 42 days. The recorded productive indices were final body weight, daily weight gain, feed intake, and feed conversion ratio (FCR). Carcass yield and percentages of liver, heart, abdominal fat, and gizzard were measured after the slaughter of representative birds. Intestinal contents were sampled to enumerate &lt;i&gt;Escherichia coli&lt;/i&gt; and &lt;i&gt;Lactobacillus&lt;/i&gt; spp., and gut pH was determined with a digital pH meter. Oxidative status and serum biochemistry, &lt;i&gt;viz&lt;/i&gt;. malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx), as well as total cholesterol (TC), triglycerides (TG), high‑ and low‑density lipoproteins (HDL, LDL), and liver enzymes AST and ALT, were assessed using commercial kits. For morphometric analysis, hematoxylin&amp;ndash;eosin‑stained sections of the duodenum and ileum were prepared to measure villus height, crypt depth, and the villus‑to‑crypt ratio. Data were analyzed in SAS software, and means were separated by Duncan&amp;rsquo;s test at p &amp;le; 0.05.&lt;/span&gt;&lt;/span&gt;&lt;br&gt;

The &lt;i&gt;S. cerevisiae&lt;/i&gt; yeast likewise decreased &lt;i&gt;E. coli&lt;/i&gt; and elevated &lt;i&gt;Lactobacillus&lt;/i&gt; relative to the control, although the magnitude of this change was lower than that observed with 2% purslane. Regarding carcass characteristics, neither purslane nor the yeast affected carcass percentage or most individual organs; however, liver percentage declined significantly in the 2% purslane group, suggesting a potential alleviation of lipid load and improved metabolic status. Intestinal pH decreased in the additive groups, with the greatest drop seen in birds receiving the yeast, an environment conducive to acid‑tolerant beneficial bacteria, such as lactobacilli. Systemically, the 2% purslane treatment significantly lowered MDA and increased the activities of SOD and GPx, indicating strengthened antioxidant defenses and reduced lipid peroxidation. The same treatment improved the lipid profile: TC, TG, and LDL were reduced compared to an increase in HDL. Histological evaluation showed that dietary purslane at 1% and 2% significantly enhanced villus height and the villus‑to‑crypt ratio and reduced crypt depth in both the duodenum and ileum. These changes are consistent with the enlarged absorptive surface area and improved digestive efficiency. The yeast also exerted favorable effects on these morphometric variables, though generally to a lesser extent than 2% purslane. Collectively, both additives modulated the gut milieu toward beneficial populations and structural optimization, but purslane at 2% achieved the most comprehensive improvements across microbial, morphological, and biochemical endpoints. The suite of bioactive constituents present in purslane seed&amp;mdash;omega‑3 fatty acids together with flavonoids and other polyphenols&amp;mdash;likely underlies the observed functional benefits. Omega‑3s can temper inflammatory pathways and support membrane integrity, while phenolic antioxidants scavenge reactive oxygen species, thereby lowering oxidative stress and protecting the intestinal epithelium. In parallel, a reduction in luminal pH and the associated increase in short‑chain organic acids favor the proliferation of commensal lactobacilli and suppress opportunistic pathogens, such as &lt;i&gt;E. coli&lt;/i&gt;, promoting colonization resistance and a more stable microbiota. &lt;i&gt;S. cerevisiae&lt;/i&gt; may complement these actions by providing growth factors and metabolites that stimulate beneficial bacteria, improving nutrient assimilation, and indirectly enhancing digestive enzyme activities. Nonetheless, in the present experiment, the breadth and intensity of responses favored the 2% purslane inclusion over the yeast alone, suggesting that the phytochemical matrix of purslane seeds confers multi‑targeted effects that extend beyond microbiota modulation.&lt;/span&gt;&lt;/span&gt;&lt;br&gt;

These convergent changes translated into a more favorable gut environment and likely more efficient digestion and absorption, thereby supporting general health in Japanese quails. The &lt;i&gt;S. cerevisiae&lt;/i&gt; yeast also produced beneficial shifts in gut flora and reduced pH, but its impacts on certain morphometric and biochemical metrics were less pronounced than those achieved with 2% purslane. Purslane seed powder, therefore, emerges as a practical phytogenic feed additive to enhance health and productivity in quails while aligning with strategies to curtail antibiotic dependence in poultry systems. Future work should examine higher inclusion levels and combined use with yeasts, as well as longer‑term monitoring of performance and immune markers, to define dose&amp;ndash;response relationships and the durability of benefits.&lt;/span&gt;&lt;/span&gt;&lt;/span&gt;&lt;/span&gt;&lt;/span&gt;&lt;/div&gt;</abstract>
