در این تحقیق چندشکلی‌های­ آللی در ژن­های کاندید TLR4 و IL2 دخیل در سیستم ایمنی برخی از نژادهای مرغ­ بومی با استفاده از تکنیک PCR-RFLP بررسی شد. 200 قطعه مرغ شامل نژاد­های مرغ بومی عمومی، آذربایجان غربی، مرندی و مازندرانی انتخاب شدند. برای شناسایی جهش در ژن­ های TLR4 و IL2 به ترتیب از آنزیم ­های Sau96I و HphI استفاده شد. برای جایگاه ژنی 257 جفت بازی TLR4، بعد از هضم آنزیمی سه ژنوتیپ CC، CG و GG شناسایی و برای آلل C دو باند 138 و 119 جفت بازی و برای آلل G سه باند 119، 99 و 39 جفت بازی مشاهده شد. در جایگاه ژنی 600 جفت بازی IL2، سه ژنوتیپAA ، AB و BB شناسایی و برای آلل A چهار باند 465، 64، 40 و 31 جفت بازی و برای آلل B پنج باند 454، 64، 40، 31 و 11 جفت بازی مشاهده شد. کل جمعیت­ ها از نظر شاخص تعادل برای دو جایگاه ژنی مورد مطالعه در تعادل هاردی - واینبرگ قرار داشت. شاخص اطلاعات شانون در جایگاه ­های نشانگری TLR4 و IL2 به ترتیب 56/0 و 69/0 و همچنین شاخص تثبیت به ترتیب 05/0- و 10/0- محاسبه شد. بیشترین مقدار شاخص هتروزیگوسیتی مشاهده شده برای جایگاه­ های ژنی TLR4 و IL2 به ترتیب 55/0 و 40/0 برآورد شد. با توجه به چندشکلی‌های موجود در جایگاه ­های ژنی مطالعه شده و کاهش هتروزیگوسیتی در این جمعیت­ ها، می ­توان از بروز تلاقی ­های غیر تصادفی جلوگیری کرد. این امر منجر به افزایش هتروزیگوسیتی و در نتیجه مانع از کاهش تنوع ژنتیکی در جمعیت­ ها خواهد شد. همچنین با مطالعه­ ی پاسخ­ های ایمنی مرتبط با این دو جایگاه ژنی می ­توان از این جایگاه به ­عنوان نشانگرهای مناسب در برنامه­ های اصلاح نژاد مرغ­ های بومی برای افزایش مقاومت به بیماری­ ها بهره برد.
