Introduction and purpose: One of the goals of identifying gene locations related to quantitative traits is to find the location of genes that affect economic traits related to growth, in this case, identifying genes with major effects on these traits can be of particular importance. in the progress of the eugenics program.The association of Ghrelin receptor gene (GHSR) with morphological and growth traits in Lori-Bakhtiari sheep breed were studied using PCR-SSCP and DNA sequencing.
Material and Methods: Blood samples were collected from jugular vein of 147 Lori-Bakhtiari sheep. Genomic DNA was extracted using phenols chloroform method. The exon 2 (452 bp) of GHSR gene was amplified using designed specific primers. The single stranded conformation polymorphism (SSCP) was used to identify polymorphism on the patterns of PCR product using 12% Acrylamide gel and silver staining method.
Results: After sequencing, one novel single nucleotide polymorphisms (silent substitution) identified in position 155 which changed nucleotide T to C. The association of identified SNP with birth weight and the upper wide fat-tail in Lori-Bakhtiari, was significant (p<0.05) and the CC genotype was higher effect than CT genotype. The studied traits CC genotype was more desirable than CT genotype.
Conclusion: The results showed that the gherlin receptor gene (GHSR) could be a possible candidate in birth weight trait for using selection in Lori-Bakhtiari sheep as a genetic marker.
