چکیده مبسوط
مقدمه و هدف: فیتواستروژن‌ها، استروژن‌های گیاهی با عملکرد و ساختار استروژن هستند. استروژن در فرآیند سنتز پروتئین‌های سفیده تخم‌مرغ در مجرای اویداکت مرغ نقش مهمی ایفا می‌کند. گیاه پنج‌انگشت یک گیاه بومی کشورهای آسیای میانه، مرکزی، جنوب اروپا و کشورهای مدیترانه است. این گیاه در طب سنتی کاربردهای متعددی دارد. مطالعات قبلی نشان داده است که گیاه پنج‌انگشت حاوی سطوح بالایی از فیتواستروژن است. فعال‌ترین فیتواستروژن‌ها در پنج‌انگشت شامل آپیژنین، وایتکسین و پندولتین است. تاکنون مطالعه‌ای در مورد اثر فیتواستروژن های گیاه پنج‌انگشت بر بیان نشانگرهای ژنی اوویداکت (اووآلبومین و اووموئید) مرغ تخمگذار منتشر نشده است. از این رو، این مطالعه به منظور بررسی تأثیر میوه‌های گیاه پنج‌انگشت بر بیان ژن های اووآلبومین و اووموئید اویداکت مرغ‌های تخم گذار انجام شد.
مواد و روش­ ها: تعداد 90 قطعه مرغ تخم گذار لگهورن (Hy-Line, W-36) در چرخه دوم تولید (در سن 72 تا 80 هفتگی) در قالب طرح کاملاً تصادفی با سه تیمار، پنج تکرار و 6 قطعه مرغ در هر تکرار مورد استفاده قرار گرفت. تیمارها شامل سطوح مختلف پودر میوه پنج‌انگشت شامل سطوح صفر، 1 و 2 درصد پودر میوه پنج‌انگشت به ازای هر کیلوگرم جیره بودند. پس از استخراج RNAی کل، کیفیت RNA با استفاده از نانودراپ و الکتروفورز ژل آگارز مورد بررسی قرار گرفت و با استفاده از کیت سیناکلون cDNA سنتز شد. میزان بیان ژن با استفاده از تکنیک واکنش زنجیره ای پلیمراز در زمان واقعی اندازه گیری شد. در این روش از ژن بتااکتین به عنوان ژن مرجع جهت نرمال نمودن داده‌ها استفاده شد. برای تایید اختصاصیت هر پرایمر نمودار ذوب آن مورد بررسی قرار گرفت.
یافته­ ها: الکتروفورز ژل آگارز محصولات واکنش زنجیره‌ای پلیمراز به ترتیب قطعات 60، 133 و 150 جفت باز را برای اووآلبومین، اووموئید و بتااکتین نشان داد. همه نمودارهای ذوب دارای یک پیک بودند که نشان دهنده حداقل دایمر پرایمر و اختصاصیت پرایمرها برای هر ژن بود. نتایج واکنش زنجیره‌ای پلیمراز در زمان واقعی نشان داد که مکمل ‌سازی میوه‌ پنج‌انگشت در سطوح 1 و 2 به جیره مرغ‌های تخمگذار تأثیری بر بیان ژن‌های اووآلبومین و اووموئید مرغ‌ تخمگذار ندارد.
نتیجه­ گیری: انتظار نمی‌رود که افزودن پودر میوه‌ پنج‌انگشت به جیره مرغ‌های تخمگذار سبب افزایش تولید در مرغ تخمگذار شود. بنابراین افزودن گیاه پنج‌انگشت به جیره مرغ های تخمگذار پیشنهاد نمی شود.
