چکیده مبسوط
مقدمه و هدف: بورخولدریا مالئی، عامل اتیولوژیک بیماری مشمشه است. تشخیص بالینی و باکتری‌شناسی مشمشه در مراحل اولیه بیماری دشوار است. برخی از روش‌ها مانند آزمایش ثبوت مکمل به دلیل سختی آزمایش و نتایج مثبت کاذب برای متخصصان دامپزشکی دردسرساز شده و موجب ضرر و زیان مالی صاحبان حیوانات می‌گردد. یکی دیگر از روش‌های تشخیصی آزمایش مالئیناسیون است که به پرسنل آموزش دیده نیاز دارد. بنابراین‌، برای تشخیص سریع و دقیق بیماری‌، به ویژه در مناطقی که نمی‌توان از حیوانات نگهداری کرد‌، باید از روش‌های جدید برای شناسایی بیماری استفاده کرد. وسترن بلات یک آزمایش تشخیصی سرولوژی است و توسط سازمان جهانی بهداشت حیوانات  توصیه شده است. هدف از این مطالعه، طراحی روش وسترن بلات با استفاده از لیپوپلی ساکارید حاوی آنتی‌ژن بورخولدریا مالئی، بود.
مواد و روش­ ها: در این مطالعه، تعداد 75 سرم از جمعیت مختلف اسب‌ها از چندین منطقه جغرافیایی ایران جمع‌ آوری شد. ویژگی و حساسیت تست‌های آزمایش ثبوت مکمل، الایزا و وسترن بلات برای تشخیص مششمه با استفاده از سرم‌های تهیه شده، مقایسه گردید. آزمایش الایزا بر‌اساس آنتی ژن‌های بورخولدریا مالئی و وسترن بلات با استفاده از آنتی ژن خالص شده بورخولدریا مالئی حاوی لیپوپلی ساکارید انجام گرفت.
یافته‌ها: نتایج نشان داد که آزمون وسترن بلات و آزمون الایزا ویژگی بالاتری نسبت به آزمایش ثبوت مکمل  داشتند. حساسیت روش الایزا بر اساس آنتی ژن‌های بورخولدریا مالئی در مقایسه با آزمایش ثبوت مکمل  و آزمون وسترن بلات بالاتر بود. در نهایت حساسیت‌ و ویژگی به ترتیب‌ برای آزمایش ثبوت مکمل  (92/31 % و 98/38%)، وسترن بلات (92/31 % ، %100)  و الایزا (%100 و 100%) بدست آمد.
نتیجه‌گیری: آزمایش ثبوت مکمل برای مشمشه هنوز روش تجویز شده سرولوژیکی برای اهداف تجاری به منظور تأیید سلامت حیوانات از نظر بیماری است. اما به دلیل حفظ امنیت زیستی کلنی‌های ارزشمند و گران قیمت اسب که پرورش آنها توسط اسب داران حرفه‌ای در مناطق مختلف ایران رو به توسعه می‌باشد بر اهمیت اعمال دقیق‌تر و هوشمندانه برنامه کنترل و ریشه‌کنی بیماری و ارتقاء توانایی‌های آزمایشگاهی تشخیص بیماری می‌افزاید تا امکان شناخت بهتر عامل بیماری و استفاده و بهینه سازی روش‌های تشخیصی مشمشه در کشور فراهم گردد. بنابراین‌، تلاش برای بهبود و بهینه سازی بیشتر الایزا و وسترن بلات می بایست ادامه یابد.
