   امروزه مشکلات تولید مثلی یکی از نگرانی­های اصلی در صنعت گاو شیری است. هدف از پژوهش حاضر شناسایی جهش فقدان عملکرد p.Q579X موثر بر سقط جنین خود به خودی و مشکلات تولید مثلی در ژن APAF1 با روش توالی ­یابی در گاو هلشتاین بود.  در مجموع 200 نمونه خون تهیه و استخراج DNA با کیت تجاری انجام شد. سپس یک جفت آغازگر اختصاصی برای تکثیر قطعه ­ای با طول 456 جفت باز حاوی جهش مورد نظر طراحی و تعیین ژنوتیپ نمونه ها با روش PCR-SSCP انجام شد. در جایگاه APAF1 دو آلل A و B و دو ژنوتیپ AA و AB به ­ترتیب با فراوانی ­های 96/76، 3/24، 93/51 و 6/49 درصد مشاهده شدند. ژنوتیپ BB در نمونه­ های مورد مطالعه مشاهده نشد. بعد از تعیین ژنوتیپ نمونه­ ها، از هر ژنوتیپ دو نمونه به­ صورت دو طرفه توالی­یابی­شده و بعد از هم­ترازی توالی­ های به­ دست­ آمده،  مقایسه آن­ها با توالی رفرنس برای شناسایی جهش­ های مورد نظر با استفاده از نرم ­افزار BioEdit (version7.0.9.0 ) صورت گرفت. نتایج بررسی­ های بیوانفورماتیک نشان داد که جهش p.Q579X که یکی از جهش ­های موثر بر سقط جنین در گاو هلشتاین می­ باشد در ژنوتیپ AB ژن APAF1 در گاوهای هلشتاین در ایران وجود دارد. با توجه به اینکه این جهش در حالت هموزیگوت باعث سقط جنین در گاو می­شود، می ­توان نتایج حاصل ­ از این پژوهش را در برنامه ­های اصلاح نژادی در راستای شناسایی حاملین و حذف آن­ها از گله گاوهای شیری هلشتاین به­ کار برد.
